What a longevity trial needs to measure

A trial’s most exciting result still needs an outcome, a comparison and a record of harms.

Find the primary outcome

Before reading which results were positive, find the measure identified as the main outcome. Was it survival, function, a diagnosis or a biomarker? The ARRIVE reporting guidance asks animal-study authors to identify outcome measures, including a primary measure for hypothesis-testing work. The point is to know what question the study was designed to answer.

Identify the comparison

A trial needs a clear account of which animals received which intervention and how that decision was made. Random allocation aims to reduce systematic baseline differences; blinding aims to reduce the influence of expectations on treatment or measurement. Neither label replaces a description of what actually happened.

The Dog Aging Project’s TRIAD key-facts document describes a double-blind, placebo-controlled rapamycin trial. That is evidence about its stated design. It is not proof that the treatment extends life, a current recruitment notice or a prescription for owners to follow.

Follow every animal through the report

Look for withdrawals, missed measurements and exclusions, with reasons. A result based only on animals completing every visit can answer a different question from one considering the original assigned groups. Read how the authors handled the difference.

Read harms beside benefits

A useful care decision needs more than a favorable change in one score. Ask what adverse events were monitored, how they were collected and how long follow-up lasted. A small or short trial may leave uncommon or delayed harms uncertain.

Keep the conclusion proportional

If the trial measured a marker, describe a marker result. If survival was not established, do not add extra years to the conclusion. A controlled experiment can move a question forward while leaving clinical practice unchanged. Decisions about a dog’s treatment belong with a veterinarian who can assess the actual animal and the available evidence.
